The identity of clinical test samples has to be verified regularly (placebo – verum). This task requires not only a fast but above all a nondestructive method. Without doubt FT-NIR transmission spectrocopy is the preferred technology for the analysis of intact tablets and BÜCHI Labortechnik AG has been identified as a competent provider and partner. The NIR experiments led not only to a validated method that could differentiate between the active ingredient and the placebo, but also yielded the prospect of the ability to discriminate between the individual strengths. An emerging task was the need to transfer the calibration from an earlier BUCHI instrument (NIRTab) to the current NIRFlex N-500 model.

The acid resistant Mini Spray Dryer B-290 version is made to spray dry acid solutions in open mode configuration. It comprises a highly effective perfluoroalkoxy polymer coating (PFA) on the metallic parts of the spray dryer which are in contact with the product and drying gas. The coating prevents corrosion and results a good quality similar to polytetrafluoroethylene (PTFE).

The Mini Spray Dryer B-290 from Büchi Labortechnik AG is a laboratory scale instrument to perform spray drying processes down to 30 mL batch volume and up to 1 litre of water or organic solvent per hour. Thanks to the glassware, the complete drying process from the two-fluid nozzle down to the powder collection vessel is visible. Fine particles are produced because of the short residence time in such a compact spray dryer. The residence time of the drying air within the spray chamber is about 1.5 seconds. The powder collection is provided by a glass-made cyclone separator, which is internally coated with a thin nanosize antistatic film to reduce powder adhesion to the glass wall. The separation works by centrifugal forces by virtue of iner tia of the solid particles.

How to Achieve Low Detection and Quantification Limits for the Nitrogen Determination with Kjeldahl

Kjeldahl is one of the most commonly used techniques to determine the protein content in food and feed samples. The detection and quantification limits are important characteristics of analytical methods. The impact of the concentration of boric acid, the addition of potassium chloride, and the concentration of the titration solution on the detection and quantification limits were investigated. The best results were obtained by using 2 % boric acid with 3 g potassium chloride per liter. A titration solution of 0.005 M HCl worked best. With these parameters, detection limits for distillation of standard solutions as low as 0.008 mg nitrogen and quantification limits of 0.02 mg nitrogen can be achieved.

Determination of PBDEs in Sediment Samples using Pressurized Solvent Extraction and Automated Soxhlet Extraction

The efficiency of pressurized solvent extraction (PSE) was investigated by comparing PSE with automated Soxhlet extraction. Sediment samples containing polybrominated diphenylethers (PBDEs) were processed in the Speed Extractor E-914 for PSE and in the Extraction System B-811 for automated Soxhlet extraction. Quantification of PBDEs was performed by GC-MS. This study shows that both techniques are comparable in terms of accuracy recovery and reproducibility and confirms that PSE is a much faster technique consuming much less solvent than automated Soxhlet extraction.

Trace Analysis of PAHs using the Extraction System B-811

The Extraction System B-811 and the Syncore Analyst parallel concentrator from Buchi as well as the Pressurized Solvent Extractor from Applied Separations were evaluated for the pretreatment of samples containing PAHs. Recoveries of extraction and subsequent concentration using acetone+n-hexane (1+1) were 70-95% for 2 ring and 90-100% for the less volatile 3-5 ring PAHs. A good precision (95% confidence level) of less than 6%, naphtalene excluded (12%), was found. No significant difference was observed between the two extraction methods.
